The trace metal working stock solution is made by dissolving 10g of ferric sequestrene in
distilled water with 1 ml of each trace metal primary stock and bringing to 1 liter

The vitamin stock solution is made by adding 1.0 ml of biotin primary stock and 1.0 ml of B
12
primary stock to distilled water with 20 mg thiamine HCL and bringing to 100 ml. All stock
solutions are kept refrigerated.
Medium f/2 is made by adding 0.5 ml of each working stock solution per liter of seawater (use
7.5 ml for 15 L carboy cultures; 30 ml for 60 L aquaculture tubes). For flask cultures nutrients
are added before autoclaving. For large cultures nutrients are autoclaved and added to the 0.2 µm
filtered seawater. Silicate forms a precipitate when autoclaved with other nutrients. If silicate is
required, it should be autoclaved separately.
7.5. Cleaning
Wear lab coat, acid gloves and safety glasses. Keep acid bath in the hood if possible. Clean up
spills immediately. Note the location of the eye-body wash station.
All filtering gear is fresh water rinsed 3 times, acid washed (10% HCl), DI rinsed 3 times, and
then air dried.
